Birmingham New Road will close from Monday October 28 until Saturday November 2, between 8pm and 5am each night.
The closure will be from the junction of Sedgley Road West to the junction of Sangwin Road.
The diversion will be via: Sedgley Road West, Hurst Lane, Bloomfield Road, High Street, Wednesbury Oak Road, Bilston Road, Great Bridge Road, Oxford Street, Black Country Route, Birmingham New Road and vice versa.
A spokesperson for Dudley Council said: "The restriction is required to safely facilitate carriageway resurfacing."
